Tadalafil (Generic Cialis) is an oral medication used to treat impotency. Tadalafil is a phosphodiesterase inhibitor which treats erectile dysfunction. The drug can be effective for up to 36 hours after consumption. Cialis is the third drug developed to cope with erectile dysfunction. The mechanism of its functioning is alike it predecessor Viagra and Levitra. It increases the volume of cyclic cGMP that will further increase the blood flow within the tissues of the male organ. This phenomenon does enable a stimulus for proper erection to be attained whenever required. It is a fact that approximately 50% of men in the age group above 40 years are experiencing erectile dysfunction. The majority of men are doing something about it. Erectile problems are not always the result of fatigue or stress. There are physical factors which can be the gearing causes; such as health conditions or side-effects experienced by other type of drugs. Tadalafil pills are yellow in colour and are shaped as almonds. The tablets of Cialis come in 3 volumes of dosages; 5mg, 10mg and 20mg. The general dosage of Tadalafil will depend upon individual tolerance to the drug. Generally, the dosage starts with 10mg and it is to be consumed prior to any sexual intercourse. The intake is then adjusted according to the patient. This anti-erectile dysfunction agent should at most be consumed once in a day. It is definitely not recommended to over consume the drug as it is said that the drug can be effective for approximately 1 ½ days. This has made the agent famously known as a “weekend Pill” thanks to its longevity.

History of Tadalafil

Tadalafil is the third erectile drug officially approved for sales in the United State. The first two drugs was Viagra and Levitra, scientifically know as sildenafil citrate and vardenafil. The FDA (Food and Drug Administration) approved the drug for sales in 21st December, 2003. It was approximately launched 5 years after the first erectile dysfunction drug, Viagra, and 4 month subsequent to the second one, Levitra.
This drug was created by Glaxo Wellcome which is today known as GlaxoSmithKline. An official agreement between Glaxo and ICOS was formed in the mid-1991 to develop a new drug. The biotechnology corporation was initiating a study on a compound known as IC351, which is a PDE-5 enzyme inhibitor. The IC351 was then developed for testing; phase 1 trial to test the compound was carried forward in 1995. The first phase was successfully completed. The Second Phase of clinical trial began thereafter to physically test the compound on erectile dysfunction. The third phase was the hallmark that concretized FDA’s approval.

The first producer Tadalafil

The partnership that prevailed between Glaxo and ICOS was dropped in 1996. The main reason that caused the lapse was diversification of business activities from Glaxo’s core market. The Lilly ICOS, LLC, Company was formed subsequently and they marketed the product Tadalafil as a special treatment for erectile dysfunction. Prior to its launch, 2002, Cialis was actually proven to have several advantages over the previous ED treatments. A clinical testing actually showed that Cialis maintained its strength for approximately 36 hours. The other two drugs Levitra and Viagra were estimated to be less than half as effective. The drug was officially released thereafter, 2003 as ICOS Corporation was taken over by Eli Lilly and Company, who bought it for 2.3 billion in 2007. Tadalafil became their property and remains traded under Cialis. The drug is today, sold as a drug used to cope with both treatment of ED and pulmonary arterial hypertension.

Mechanism of Tadalafil

Tadalafil, drug is used to cope with problems concerning the sustainability of penile erections. This drug is in the same group as Sildenafil and Vardenafil which is called PDE-5 inhibitors. An erection is generated by increasing blood pressure to a sufficient amount capable of withholding an erection. The flow of blood must be effectively propelled through blood vessels. The further increase in blood flow, the greater will the physical increase of the penis be.

Tadalafil is a drug that can improve the flow of blood in the veins of the penis. It is acknowledged that sexual stimulation releases nitric oxide which will initiate the process. The nitric oxide (NO) will root an enzyme to produce cGMP. The cGMP is the main element responsible for the regulation of blood in the arteries carrying blood to and away from the penis. The male organ will be erected as long as the cGMP is not wiped out by another enzyme, PDE-5. Once this occurs the blood will retaliate and the state of the penis will become normal (not erected). Tadalafil has a special function of preventing PDE-5 to intervene in the process of destroying the cGMP. This does ultimately allow a prolonged penile erection. The male organ will remain engorged for as long as the blood remains present in the arteries of the organ.

Tadalafil does not directly affect the state of the organ. The presence of sexual stimulation is of essence to enlarging its size. It was highlighted by the European Commission, 2007, that a dosage of about 5mg to 2.5 mg can be consumed at daily basis. The drug is at the moment being tested to treat other types of health issues such as pulmonary arterial hypertension. It has been successful at different phases and it was shown in phase 3 that 40mg of Tadalafil would be the most appropriate dose to deal with pulmonary arterial hypertension as communicated through a conference in 2008.

Preparation and Precaution while using Tadalafil

Cialis should only be provided to a patient by the prescription of a doctor. A healthcare provider will be able to Tadalafilascertain that the product is compatible with the individuals’ actual conditions. The degree of tolerance and reaction to drugs such as tadalafil differ among users. There is a wide range of users who are only prescribed for a low intake of the drug. The healthcare provider will prescribe two types of tadalafil. The first one is a 36-hour Cialis and the other is Cialis for daily usage.

The 36-hours Cialis should not be taken more than once per day. It is recommended that the dose of tadalafil is consumed at least 30 minutes prior to any sexual intercourse. The drug will remain active and effective up to 36 hours. The medication will lead to an erection, only after sexual stimulation has been initiated. “Cialis for daily dosage” (second pill) should also be taken only once daily. It will function equally as the 36-hour pill. The type of tadalafil pill will depend upon the healthcare provider.

It is strictly advised not to use more than one ED medicine. Tadalafil is not necessarily compatible to be consumed simultaneously with any other PDE-5 inhibitors. The degree of alcohol consumed can also be the general cause towards side-effects such as headache and dizziness. Alcohol will lower blood pressure and rise heart beat. It is thus advised not to over consume alcoholic drinks while under tadalafil medication.

Risk and Side Effects of Tadalafil (Cialis)

Tadalafil has been used by millions of people world-wide and thousands of people have participated in clinical trials. The drug is not a miracle as it does definitely contain some side-effect similarly to any other medication. The most prevalent shortcomings of Cialis (Tadalafil) have been fatigue, headache, muscle aches, backpain and stomach related issues. These are usually temporarily inconveniences that do regularly relieve themselves within a few hours and at worst within a day or two. Cialis can unfortunately lead to priapism (long lasting erection for above 4 hours) which is prevalent in any kind of erectile medication. It is painful but extremely rare.

The PDE-5 inhibitors such as tadalafil have been alerted, since 2007, that they can have a significant relationship with deafness. The coherence of the prominent effect of hearing has indeed not been fully analysed and the extent of the relationship between PDE-5 inhibitors are still uncertain as announced by FDA.
